On the Quanta of Light Produced and the Molecules of Oxygen Utilized during Cypridina Luminescence

چکیده

A study of the oxygen consumed per lumen of luminescence during oxidation of Cypridina luciferin in presence of luciferase, gives 11.4 x 10(-5) gm. oxygen per lumen or 88 molecules per quantum of lambda = 0.48micro, the maximum in the Cypridina luminescence spectrum. For reasons given in the text, the actual value is probably somewhat less than this, perhaps of the order of 6.48 x 10(-5) gm. per lumen or 50 molecules of oxygen and 100 molecules of luciferin per quantum. It is quite certain that more than 1 molecule per quantum must react. On the basis of a reaction of the type: luciferin + 1/2 O(2) = oxyluciferin + H(2)O + 54 Cal., it is calculated that the total efficiency of the luminescent process, energy in luminescence/heat of reaction, is about 1 per cent; and that a luciferin solution containing 4 per cent of dried Cypridina material should rise in temperature about 0.001 degrees C. during luminescence, and contain luciferin in approximately 0.00002 molecular concentration.

The discovery of aequorin and green fluorescent protein.

We discovered aequorin and green fluorescent protein (GFP) in 1961 from the same species of jellyfish (Shimomura et al ., 1962). Our target was a luminescent substance, aequorin, and GFP was isolated as a by-product of aequorin owing to its bright conspicuous fluorescence. Both are unusual proteins but they had no particular importance when we first reported them. Their importance became appare...

Studies on Bioluminescence. X. Carbon Dioxide Production during Luminescence of Cypridina Luciferin. by E. Newton Harvey

All luminous animals require oxygen for luminescence bu t it is not known whether they give off CO2 as a result of the luminescence. Luminous bacteria produce carbon dioxide but this is no doubt the carbon dioxide of respiration. Even if we could prove that luminous bacteria produced more carbon dioxide than certain non-luminous bacteria, this would not necessarily znean that the excess carbon ...

The Inhibition of Cypridina Luminescence by Light

The luminescence of Cypridina luciferin-luciferase solution is inhibited by illumination from a carbon arc of 15,000 foot candles in between 1 and 2 seconds. The blue to violet rays are the effective ones, the limits lying somewhere around 4,600 A. u. to 3,800 A. u. The luciferin, not the luciferase, is the substance affected by the light. The effect is partially reversible in the dark. The che...

Studies on the Oxidation of Luciferin without Luciferase and the Mechanism of Bioluminescence

In five groups of luminous animals it can be shown that luminescence is connected with the oxidation of a substance, luciferin, in aqueous solution, in presence of an enzyme, luciferase, and dissolved oxygen. In the mollusc, Pholas dactylus, Dubois (1) has been able to oxidize Pholas luciferin with luminescence by KMn04, HzO, BaOz, PbOa, and hypochlorites. I have been unable to oxidize with lum...

Studies on the Amount of Light E M I

Interest has centered for a long time on the intensity and efficiency of animal luminescence, not only from a theoretical but also from a practical standpoint. With the exception of the beetles, most luminous animals emit light in such small amounts that adark-adaptedeye is required to register their intensities. I t is a difficult matter to measure accurately low light intensities, especially ...
